What is Dimethylpropiothetin (DMPT)?

Dimethylpropiothetin (Dimethyl Beta Propiothetin, DMPT, DMSP) is a naturally occurring organosulfur compound that has gained significant attention in the aquaculture and animal nutrition industries. This compound belongs to the class of sulfur-containing compounds known as sulfonium salts. Dimethylpropiothetin is structurally characterized as a sulfonium compound containing a positively charged sulfur atom bonded to two methyl groups and a propionate group. The compound appears as a white to off-white crystalline powder at room temperature and demonstrates high solubility in water, making it particularly suitable for aquatic applications. Its stable chemical structure under normal storage conditions contributes to its practical utility in commercial applications. The compound’s unique sulfur-based structure is responsible for its distinctive properties and biological activities. The presence of the sulfonium group creates a permanent positive charge, which influences its interaction with biological systems and its ability to function as an effective feed attractant. In the EU, DMPT is permitted as a feed attractant for fish and shrimp, but only in hook baits (lures) according to the EU regulation 1831/2003. DMPT HCl salt also has the same applications.

Natural Occurrence and Biological Significance

DMPT occurs naturally in various marine organisms, particularly in algae, seaweeds, and certain marine animals. Marine algae synthesize DMPT as part of their normal metabolic processes, where it serves multiple functions, including osmoregulation and stress response. This natural occurrence explains why many aquatic animals have evolved receptors sensitive to DMPT and similar compounds. In marine ecosystems, DMPT plays a role in chemical communication between organisms. When released into the water, it can serve as a chemical signal that influences feeding behavior and other biological responses in marine animals. This natural relationship between DMPT and marine life forms the basis for its commercial applications.

Applications in Aquaculture

DMPT, as a feed attractant for aquatic animals, significantly promotes the feeding behavior of many freshwater and marine fish, shrimp, and crabs. As same as IMP and TMAO, DMPT can increase the bait-biting times of aquatic animals, and its attracting effect is 2.55 times higher than L-glutamine (L-glutamine was the best-known feeding stimulant for most freshwater fish before DMPT). Research has demonstrated that DMPT supplementation can increase feed intake by 20-40% in various fish species, including carp, tilapia, and marine fish. This enhanced feed consumption translates to improved growth rates and feed conversion efficiency, making it economically valuable for aquaculture operations. The compound is particularly useful during stressful periods such as environmental changes, disease treatment, or when introducing new feed formulations.

Mechanism of Action

DMPT functions through multiple mechanisms to enhance feeding behavior and growth performance. The primary mechanism involves the stimulation of olfactory and gustatory receptors in aquatic animals. The sense of aquatic animals can be stimulated by low concentrations of attractants in the water. Therefore, fish, shrimps, and crabs have a strong physiological mechanism of attracting food with the special smell of DMPT.

Safety and Regulatory Considerations

DMPT has undergone extensive safety evaluations and is generally recognized as safe for use in animal feeds when used in accordance with recommended guidelines. Regulatory authorities in major aquaculture-producing countries have approved its use as a feed additive. The compound demonstrates low toxicity to aquatic animals, with effective doses well below levels that might cause adverse effects.

Manufacturing Process of Dimethylpropiothetin (DMPT) Fish Attractant

Natural Extraction Process

The earliest DMPT production involved extracting the compound directly from marine algae using water as the primary extractant. This natural extraction process represented the foundation for understanding DMPT’s chemical properties and biological activity. Marine algae, particularly species with high DMPT concentrations, served as the raw material for this extraction method. The natural extraction process, while providing authentic DMPT compounds, suffered from several inherent limitations. The variable concentration of DMPT in different algae species, seasonal fluctuations in algae availability, and the complex purification requirements made this method economically unfeasible for large-scale production. Additionally, the relatively low purity of the extracted compound limited its effectiveness in commercial applications.

Chemical Synthesis Process

Recognition of the limitations associated with natural extraction led to the development of synthetic manufacturing processes. Chemical synthesis offered several advantages, including consistent product quality, higher purity levels, reduced production costs, and the ability to scale production according to market demand.

  • Step 1: Clean the 2000L enamel reactor, turn on the vacuum water pump, open the vacuum valve and feeding valve on the reactor, close other valves, accurately weigh 150kg of acrylic acid, 320kg of water, and 330kg of hydrochloric acid in turn, pump them into the reactor and stir, and open the drain valve on the reactor.
  • Step 2: Cool the reactor to 2℃ and stir evenly.
  • Step 3: Use the feeding pump to add 150kg of dimethyl sulfide to the reactor, observe the temperature in the reactor during the addition process, and control the flow rate so that the temperature in the reactor does not exceed 50℃.
  • Step 4: After the addition is completed, open the reflux system valve, raise the reactor temperature to 50℃, and keep it warm for 1 hour. Then raise the reactor temperature to 60℃ and keep it warm for 1 hour; then raise the reactor temperature to 75℃ and keep it warm for 1 hour.
  • Step 5. After the reaction is over, if there is residual sulfide in the U-tube, put the residual sulfide into the raw material barrel of sulfide and use it as raw material; turn on the vacuum pump, open the vacuum distillation system valve, close other valves, decompress water, control the liquid temperature in the reactor to not exceed 70℃, and distill 500kg of water. At the same time, observe the solid precipitation in the reactor through the feeding port sight glass. The distillation is over, and the temperature is lowered.
  • Step 6. Cool down to below 60℃, and use the vacuum in the reactor to draw in 500kg of acetone. When the vacuum is insufficient, open the reactor drain valve and use the feed pump to pump the remaining acetone into the reactor; after the feeding is completed, continue to cool down to below 20℃, discharge the material, centrifuge, dry, the drying temperature is 55℃, dry for 5 hours, take samples to test the drying loss is less than 1%, take out of the oven, crush and package.

The successful development of synthetic DMPT manufacturing has enabled the widespread adoption of this highly effective feeding stimulant across various aquatic applications. Continued refinement of manufacturing processes and quality control measures ensures that DMPT remains a valuable tool for enhancing aquatic animal nutrition and fishing effectiveness while maintaining environmental responsibility and production sustainability.

Dimethylpropiothetin (DMPT) Used in Fish Bait

A Freshwater Fish Bait Containing An Attractant

Includes the following steps:

Step 1. Fish Attractant Preparation

Amino acid attractants, betaine attractants, sulfur-containing compound attractants, fat attractants, and small peptide attractants are crushed and wet-mixed in a ratio of 1:3:1:2:3, and then water is added in small amounts and multiple times during the process to ensure that the overall humidity is maintained at 30% to form a mixed attractant.

  • Amino acid attractants use L-Glutamine, which is considered to be one of the most effective compounds. L-Glutamine has a strong stimulating effect on the smell and taste of fish.
  • Betaine attractants use betaine extracted from byproducts of beet processing. Betaine can increase fish feed intake, shorten feeding time, reduce the feed coefficient, and promote fish growth.
  • Sulfur-containing attractants use dimethylpropiothetin DMPT, dimethyl sulfoxide, and allicin, with a ratio of 3:2:5. Sulfur-containing compound attractants combined with basic fish baits have different degrees of promoting effects on the growth, feeding, and stress resistance of various freshwater fish.
  • Fat attractants use phospholipids derived from animal and plant fats. In addition to providing glycerol, fatty acids, phosphoric acid, choline, and inositol in animals, phospholipids can also promote the absorption of lipids in baits.
  • Small peptide attractants use attractant peptides, nucleotides, soluble fish protein, and taurine, with a ratio of 2:3:1:4. Small peptide attractants increase the feeding frequency of fish, increase the growth rate and survival rate of fish, not only stimulate the appetite of fish, but also quickly attract fish to approach.

Step 2. Stick Them in Layers

Stick the attractant, basic fish bait and absorbent floating bait in a three-layer structure of attractant layer, fish bait layer and absorbent floating layer; mix bean dregs, peanut dregs, vegetable dregs and rice bran in a ratio of 2:3:2:3 to form absorbent floating bait; mix the attractant, fish bait and absorbent floating bait in a ratio of 1:8:1 and stick them in layers. The center is a water-absorbing floating layer made of water-absorbing floating bait. The water-absorbing floating layer is adhered to the fish bait layer made of basic fish bait, and the fish bait layer is adhered to the attractant layer made of mixed attractants. The adhesive material between each layer is a molasses solution to ensure that each layer of bait in each portion of the bait is layered and firmly adhered.

Step 3. Extrusion Molding

The mixed bait can be formed into long strips of bait by extrusion and machine granulation to facilitate feeding and management.

Step 4. Drying and Cooling

The molded bait needs to be properly microwave-dried. The drying process takes 10 minutes at a time. It is carried out multiple times and measured in real time to ensure that the moisture of the fish bait is maintained at 10%-15% to remove excess moisture, and then cooled to a suitable temperature.

Finally, it is sealed and packaged to ensure the quality and safety of the bait.

In conclusion, dimethyl-beta-propiothetin represents a valuable ingredient in modern aquaculture, offering scientifically proven benefits for feed attracting efficiency. Its natural origin, established safety profile, and demonstrated effectiveness make it an important component of sustainable aquaculture practices. As a bulk manufacturer and supplier of dimethylpropiothetin DMPT in China, we sell different grades of fish attractant DMPT at wholesale prices to large manufacturers and small customers worldwide.
